Yes most definitely benefits (assuming the new live rock is fully cured). Diversifying bacteria is always good.

Increases biodiversity. Allows you to change up your aquascape.

Obviously make sure the new rock is cured before adding it.
 
Depending on where the new rock comes from, keep in mind you may also run the risk of introducing hitchhikers or something you don't want.
 
Are you sure it's not measurement error? A faulty or incorrectly performed test sounds more plausible than a mature tank with measurable nitrite (assuming you have a reasonable amount of rock/sand and fish load has not recently dramatically increased).
 
Hobby grade Nitrite test kits are not accurate for salt water, it has been known for years. So please don’t worry about Nitrite.
